It took just a few seconds to convert and I was able to expand the drive beyond 2TB afterward as needed. Create a new virtual drive, mount it in the VM, and then copy the files to it.
Unmount the old drive keep for backup and then remount the new drive as the old drive letter. I am still a little confused on how your virtual disk is setup but I have had a similar issue before with VMDKs. I was able to extend the virtual disk then from Windows convert the disk to dynamic and expand the drive. Since I did this on a cloned VM and not the server in use I wouldn't recommend it because too many factors could have made this not work. I would suggest using multiple smaller VMDKs.
Not sure how you have things structured if it would be amenable to doing this. I would suggest that if you don't have enough free capacity to make a copy of your largest VMDK that you need to either make that VMDK smaller, or get more free space. You are not in a good spot right now. You should consider breaking up that volume into smaller chunks to make restores that much quicker. I'm pretty sure that changing the cluster size is a destructive process. At least with the inbuilt tools.
